After the success of the 21 Jump Program, the duo of Schmidt and Jenko is put on an undercover program to work for Captain Dickson, who is located across the street at 22 Jump Street. Their assignment is to be undercover as college students to discover who supplies the drug name ‘ WHY-PHY’. This drug has already killed a student. Later in the story, Jenko becomes friends with Zook and Rooster, who were prime suspects in the investigation. After some time Jenko starts attending parties who do not prefer taking Schmidt. Meanwhile, Schmidt gets the attention of an art student, Maya, who was later discovered as the daughter of Captain Dickson. Until now, Schmidt and Jenko were unable to find the culprit, thereafter they visited Mr. Walters and Eric in the jail. In jail, Mr. Walters tells them the culprit has a unique tattoo on his arm, Jenko notices a tattoo on Zook’s arm, with whom Schmidt used to play football. Soon after they found out Zook is not the dealer. Also, they found a man and ghost on their campus. In the summer break, Schmidt goes after Ghost. While the following Ghost, Jenko got shot, later when Ghost was escaping the duo couldn’t catch him. The Ghost celebrated his victory. Afterward, Jenko said to Schmidt that they could not solve it because of their differences, so there is a need to solve the differences first, and the two reconcile.
